Quantitative LC-MS/MS can perform an important role in the development of immunoassays. We have optimized our LC-MS/MS quantitative methods by incorporating the use of labeled internal standards with sample preparation steps including solid phase and liquid/liquid extractions followed by liquid chromatography-tandem mass spectrometry multiple reaction monitoring. The LC-MS/MS analytical standard curves and controls encompass the corresponding immunoassay standard curve ranges for the analysis and standardization of numerous immunoassay analytes including immunosuppressant and oncology drugs, and fertility and metabolic hormones. These analytes are measured by LC-MS/MS in various biological matrices including whole blood, plasma, serum, and urine. These LC-MS/MS quantitative analytical method studies are essential for the confirmation of assay calibrator analyte concentration levels, clinical correlation studies, quantitation of immunoassay matrix components and identification and quantitation of analyte metabolites. Quantitative LC-MS/MS therefore provides a fundamental tool for the immunoassay development process.
