Steroidal glycosides are known to have a hypoglycemic effect in mouse models. Previously five novel steroidal glycosides were isolated and identified in bulbs of the Easter lily (Lilium longiflorum Thunb.), three are saponins and two steroidal glycoalkaloids. Two extracts were prepared from L. longiflorum bulbs: a crude freeze-dried bulb powder (Bulb) and a partially purified butanol extract (BuOH). Our initial results showed that 0.075% Bulb in the drinking water significantly lowered blood glucose in female KK CgAy/J mice (p<0.05) through increased insulin secretion. In this study, the levels of the five novel steroidal glycosides in both Bulb and BuOH extracts are quantified by LC-MS and their anti-diabetic effects evaluated and compared using the KK CgAy/J mouse model by assaying body weight, serum glucose level, serum and pancreatic insulin levels, glucose tolerance, proinflammatory cytokines in the liver, intestinal disaccharidases and histological changes in the liver and pancreas.
