The development of novel ultrasensitive methods for the detection of  biomarkers at femtomolar and even attomolar concentrations is crucial  for the development of future clinical diagnostics.  These  concentrations are typically beyond the capabilities of traditional  enzyme-linked immunosorbent assays (ELISA).   Previously in our group, surface plasmon resonance imaging (SPRI)  biosensing has been used in conjunction with surface enzymatic  amplification methods for the ultrasensitive detection DNA and microRNA  at these concentration levels. In this presentation, we describe a set  of new surface enzymatic methods for detecting protein biomarkers at  extremely low concentrations.  Surface immobilized DNA aptamer switches  are used in conjunction with RNA transcription amplification and  biofunctionalized silica nanoparticles for enhanced SPRI biomarker  detection. Further work on the on-chip synthesis of ssRNA with RNA  polymerase to create RNA aptamer microarrays for ultrasensitive protein  biosensing is also reported.
