When reversed-phase high-performance liquid chromatography (RP-HPLC) is performed at high temperature, the mobile phase used can contain little to no organic modifier (such as methanol or acetonitrile) as compared to the conventional approach thereby lending to a greener method of analysis. High temperature RP-HPLC was used in the separation of caffeine, acetaminophen and acetylsalicylic acid at temperatures from 50�C to 110�C a nd mobile phases comprised of 95 % 0.65 M aqueous acetic acid/5 % methanol or 100 % 0.65 M aqueous acetic acid. Van't Hoff plots showed good linearity as predicted from theory for all conditions employed. Results for the quantification of caffeine in an analgesic preparation containing acetaminophen and caffeine showed that high temperature RP-HPLC has the potential for use in routine pharmaceutical analysis.
