Cholesterol forms domains involved in Alzheimer's progression. Research indicates Langmuir monolayers help understand phospholipid-cholesterol interaction in forming membrane outer leaflet domains. Monolayers enable amphiphilic molecular interactions at air-water interfaces to be determined by varying area while monitoring surface pressure at constant temperature (isotherm).  Isotherms confirm that dipalmitoylphosphatidylserine (DPPS), an anionic inner leaflet lipid, forms cholesterol domains. To determine tailgroup interactions, we propose the use of bio-active cholesterol precursors. The data reveals that structural changes in the precursors' tailgroups affect molecular area. These DPPS-precursor interactions will determine if tailgroup interactions exist.  This knowledge allows for a complete picture of cellular membrane domain use in both leaflets in signal transduction for Alzheimer's.
