Chlorophyll a is an important parameter for monitoring the biological productivity of lakes. Chlorophyll a can be measured in the laboratory using spectrophotometry and HPLC with visible or fluorometric detection. Also, fluorometric field probes can be used for the in situ detection. Data obtained over several years from two highly productive lakes in Southern California show poor correlation between chlorophyll a values obtained using the standard spectrophotometric method (SM 10200H-2), and those obtained using an in situ field probe (HACH Hydrolab Chlorophyll a sensor). Hence, there is a need for a more accurate and robust method for measuring chlorophyll a in surface waters. Starting with EPA Method 447, we are optimizing the use HPLC with visible detection for measuring chlorophyll a, and the results are being statistically compared to results from both the spectrophotometric method and in situ probe for split samples collected at the same times and locations.
