Based on the high sensitivity, high specificity and high stability derived from biotin-avidin system (BAS), and the characteristics of magnetic nanoparticles ?-Fe2O3, for example, low biological toxicity, no immunogenicity, strong enzyme-resistant ability, target-based etc, the method of magnetic enrichment target sequence polymerase chain reaction (PCR) was established. We determined the best hybridization temperature of specificity bio-primer with target sequence is 53?, the optimal dosage of the magnetic nanoparticles ?-Fe2O3 with a concentration of 10 mg/mL is 15 mL. This detection limit for target fragment was performed and the minimum detection concentration is 1 pg/mL.
