Highly concentrated metals such as Cu, Zn, and Fe are found in amyloid-beta (A�) plaques within the brain of Alzheimer's disease (AD). Though the mechanism still remains unclear, in vitro and in vivo studies have suggested that metal binding to A� could facilitate A� aggregation and generate reactive oxygen species (ROS), which could contribute to the neuropathogenesis of AD. The association between metal-A� interaction/reactivity with AD development, however, has not been clearly revealed. In order to better understand this, bifunctional small molecules that can target metal-A� species and modulate their interaction/reactivity have been developed. Here, the synthesis, characterization, and reactivity of a new class of bifunctional molecules for targeting and regulating metal-A� species will be presented.
