Our interest in [Rh2(O2CCH3)2(CH3CN)6]2+ stems from the 34-fold enhancement of its cytotoxicity towards Hs-27  human skin fibroblasts upon photolysis and its binding to dsDNA. Herein,  we investigated, by 1H NMR spectroscopy, the mechanism by which 2,2�-bipyridine, a biologically relevant model, binds to [Rh2(O2CCH3)2(CH3CN)6]2+,  in the dark and upon photolysis, to form the final adduct with bpy in  eq/eq positions.  The results clearly highlight the importance of light  activation for the initial eq ligand exchange which is enhanced 20-fold  upon irradiation, a finding that hints that such dirhodium compounds are  potentially useful PDT agents.
