Drug delivery by release of a 'caged' molecule has quickly emerged as a potential therapeutic method for cancer treatment. Nitric oxide can be released photochemically from metal centered compounds and is known to increase radiation induced cell death up to 100-times relative to hypoxic controls in cancerous cells. Here is discussed the synthesis and photochemical studies of several ruthenium salen nitrosyls which undergo light induced nitric oxide release (salen2- = N,N'-ethylenebis(salicylideneiminato) dianon). These ruthenium salen compounds have been modified by hydroxide groups, methyl esters, and alkyne groups to allow for several tethering possibilities to quantum dots (QD) and homing peptides for biological studies. Quantum yield studies of nitric oxide release along with energy transfer mechanisms have also been explored.
