Flavonoids found in dietary fruits and vegetables are thought to decrease incidence of degenerative diseases, possibly through antioxidant activity. Previous work showed that flavonoid-containing polyphenolic extracts made from the foliage of Artemisia tridentata ssp. tridentata, or basin big-sagebrush, possessed in vitro antioxidant properties. Glutathione (GSH) is a small compound synthesized from amino acids which acts as a cellular antioxidant and is essential in protecting against the damaging effects of free radicals. To investigate effects of sagebrush polyphenols on GSH, HepG2 cells were treated with sagebrush extracts and the levels of GSH assayed.  Dose- and time- dependence of the effects of sagebrush on cellular GSH were analyzed to determine the effective concentration and time of treatment. Characterization of the effects of sagebrush polyphenolic extracts on cellular GSH levels will help elucidate the cellular mechanism by which these compounds exert bioactivity.
