Recognized as a disease of poverty, tuberculosis infects more than 2 billion individuals globally, and out of that inflicted population, 1.7 million individuals die as a consequence of the dire effects of this disease.  There are various forms of tuberculosis that have proven to be resistant to an array of antibiotics.  In our marine natural products lab, our emphasis is to discover novel antibiotics for tuberculosis through investigation of marine bacterial isolates.  One marine isolate, UA 461, was collected from San Elijo Lagoon in Cardiff, California in 2002.  The initial extract of  UA 461 exhibited significant inhibitory activity against Mycobacterium marinum, the closest genetic relative of M. tuberculosis, but the inhibitory activity was only present when the strain was challenged with M. marinum during culture.  On large scale culture of UA 461, solid phase extraction was used to generate three initial fractions based on polarity.  A fluorescence-based M. marinum growth assay was then used to guide purification of the active compounds using chromatographic methods, including a final step of HPLC.  NMR and other spectroscopic methods were used to identify structural characteristics.
