In this study, we focused on setting up the new detection procedure of oligonucleotide through the microarray using DNA-conjugated Pd nanoparticles (NPs) as the label. In the preliminary experiment, DNA-conjugated Pd NPs were hybridized with four kinds of ssDNA probes (complementary, one base mismatch, two base mismatch and noncomplementary mismatch) immobilized on the glass slide. After Pd Label and subsequent precipitation of silver by the electroless plating, which is similar to the procedure of Au label Ag stain presented by Mirkin C.A. et al., the results show that the signal could make the effective discrimination among the above four ssDNA probes. In comparison with the method of Au label Ag stain, this method offers a dramatic enhancement of hybridization response, and the time needed for the deposition of Ag catalyzed by Pd NPs is less.
