Pseudomonas aeruginosa bacteria contribute to cystic fibrosis (CF) patient morbidity and mortality by forming biofilms in patient lungs. Biofilms form in response to the quorum sensing (QS) molecules N-butyryl-L-homoserine lactone (C4) and N-(3-oxodo-decanoyl)-L-homoserine lactone (3OC12). If this biofilm forming process could be interrupted, CF patient prognosis could be much more positive. Extract from PAO1 cultures was separated using high performance liquid chromatography (HPLC) with a C8 reverse phase analytical column, and the identities of putative QS molecule peaks verified using proton nuclear magnetic resonance (H-NMR) spectroscopy.  Previous research indicated a significant C4 presence in the putative C4 peak.  After further analysis, the putative C4 peak was shown to actually contain two compounds. Verification of these peaks is waiting mass spectroscopy  analysis. Ultimately, this research should positively identify both C4 and 3OC12 in biological samples, and use this information to compare the production of these two molecules during biofilm formation.
