The aggregation of amyloid-beta (Ab) and its subsequent deposition as fibrils is strongly implicated as a causative factor responsible for the development of Alzheimer's Disease (AD). Preventing and reducing Ab aggregation is considered to be a critical solution in avoiding AD. Transthyretin (TTR) is a major Ab-binding protein in human cerebrospinal fluid (CSF), and may play an important role in preventing amyloid formation. Previous studies have shown that TTR can sequester Ab directly and affect Ab aggregation and toxicity, protecting against neurodegeneration via clearance of Ab Therefore, the detection of TTR in CSF might provide a possible diagnostic and therapeutic approach for AD.  Here, we used surface plasmon resonance (SPR) to detect the level of TTR in CSF in real-time and tested the hypothesis that a characteristic feature of CSF from AD patients may have reduced levels of TTR.
