We studied the structure and characteristic of depectinated primary plant cell wall from wild and double mutant xxt1xxt2 Arabidopsis separately using small-angle neutron scattering (SANS). We take the advantages of deuterium labeling and contrast match method to distinguish cellulose and hemicellulose in the depectinated plant cell walls. The dominant hemicellulose in xxt1xxt2 Arabidopsis, the arabinoxylan, is determined to exhibit a random-walk polymer behavior (theta solvent), and form a network with a mesh size (27nm to 90nm) which is identical to cellulose network in the system. Additionally, the dominant hemicellulose in the wild type, the xyloglucan, Arabidopsis also shows a random-walk behavior, but form a network with a bigger mesh size (45nm to 140nm) than what we found in xxt1xxt2 Arabidopsis.
