Our lab has recently demonstrated the ability of a split aptamer to translate the presence of a small molecule into the output of DNA ligation.  Using the DNA split aptamer for cocaine, we have achieved dose-dependent ligation over a wide range of analyte concentrations.  Additionally, we have engineered the split aptamer to function in human blood serum.  Here we report the application of this split aptamer ligation technology for development of an aptamer-based enzyme-linked assay.  This new assay functions analogously to enzyme-linked immunosorbent assay (ELISA), but utilizes a split aptamer in place of antibodies, taking advantage of the lower cost and higher chemical stability of DNA relative to antibodies.
