Abalone is a marine gastropod, massively maricultured in Asia, Africa, Australia, and America. However, health beneficial effects of the abalone have rarely been reported. AIGID I (> 100 kDa), AIGID II (100 - 10 kDa) and AIGID III (< 10 kDa) were prepared from abalone gastro-intestinal digests (AIGIDs) by a membrane ultrafiltration system. AIGID III (< 10 kDa) significantly diminished LPS-induced NO over-production and was further investigated. A peptide responsible for the anti-inflammatory activity was isolated and its sequence was identified as Pro-Phe-Asn-Glu-Gly-Thr-Phe-Ala-Ser (1175.2 Da). It was found that the purified peptide significantly exhibited the inhibitory activity on inflammatory mediators of inducible enzyme of iNOS and pro-inflammatory cytokines of TNF-a, IL-1�, and IL-6 in RAW264.7 cells. Additionally, administration of purified peptide showed significantly reduced effects of MAP kinases via the attenuation of the phosphorylated ERK-1/2 and p38 protein kinase expression in RAW264.7 cells. It was suggested that the purified peptide could be considered as a potential candidate for anti-inflammatory therapeutic substances and functional food.
