Binding of metalloproteins to self-assembled monolayer (SAM) coated surfaces is elucidated by spectroscopic investigations of cytochrome c and cytochrome cb562, which are immobilized on gold nanoparticles that are coated with alkanethiol SAMs with various functional terminating groups and varying alkane chain lengths. Steady-state absorption spectroscopy, circular dichroism and reduction potentials are employed to gain insight into the folding status of the SAM-bound cytochromes. Atomic force microscopy studies assess the amount of aggregation of cytochrome c and azurin on various SAM surfaces. These investigations are crucial for proper interpretation of cyclic voltammetry studies of metalloproteins bound to SAM coated gold electrodes to obtain electron transfer rates.
