We describe here a class of macrocyclic peptides that adopt a �-sheet structure and can present a variety of heptapeptides from amyloids in a pre-organized �-strand conformation. These amyloid �-sheet mimics (ABSMs) contain the turn and template units dOrn and Hao, which help enforce a �-strand conformation in the heptapeptide strand and block aggregation. ABSMs containing suitable heptapeptide sequences inhibit amyloid aggregation and protect cells from the toxic effects of amyloids.
