High-throughput cell-based assays are powerful tools for screening complex samples for effects on biological signaling pathways.  These �bioassays� represent an increasingly important complement to traditional analytical chemistry techniques for addressing growing concerns about the levels of pharmaceutical compounds, particularly endocrine disrupting compounds, in water supplies.  Our unique collection of 18,000 pre-cloned human transcriptional regulatory elements in the LightSwitch Luciferase Assay System represents a virtually unlimited resource for the development of bioassays.  From this set, we have paired validated pathway-specific biomarkers with a novel multiplex assay format to create the next generation of highly sensitive and reproducible bioassays for identifying samples containing activators of the estrogen, androgen, and glucocorticoid receptor pathways.  In this study, we characterized the assays and then screened a diverse panel of water samples.  The results both replicated the findings from traditional analytical chemistry screening techniques and yielded exciting new discoveries.
