Cancer is a �disease� of the cell cycle. Unlike normal cells, cancer cells have evolved biological mechanisms that enable them to evade the cell cycle check points and proliferate even if extensive DNA damage is detected. In our research, a proteomics/mass spectrometry approach was developed for characterizing the G1 and S stages of the cell cycle of MCF-7 estrogen positive breast cancer and MCF-10 normal cells. From a total of ~3700 proteins, a subset of ~600 qualified for differential expression analysis. Data interpretation was enabled by using bioinformatics tools provided by the Gene Ontology, Go Miner, DAVID and STRING software packages. A broad range of cell cycle proteins, representative of all hallmarks of cancer, were identified. Among these, of particular relevance were differentially expressed proteins implicated in protein interaction networks that span across processes that affect genome integrity such as signaling, transcription, chromatin remodeling, DNA repair and RNA splicing.
