Protein homeostasis is essential for all cells and requires the proper folding, localization and interaction of all the cellular proteins.  The signal recognition particle (SRP) is a universally conserved cellular machinery that mediates the targeted delivery of membrane and secretory proteins, and has served as a paradigm to understand the molecular basis of protein localization.  Here we describe how a series of elaborate conformational changes in the SRP and SRP receptor provide multiple regulatory points that together ensure high fidelity of substrate selection by the SRP.  We will discuss the molecular mechanisms that enable the allosteric communication between the SRP and SR and their biological cues, thus ensuring highly productive and efficient protein targeting reactions.
