Peptides that bind to a desired protein target have several features that make them attractive as molecular recognition elements in biosensor platforms and for therapeutics development. Peptides have a high structural diversity and intrinsic compatibility with the proteins that allow them to achieve high binding affinity to the target. We propose to use a combination of in silico design and microarray screening for high-throughput selection of protein binding peptides. Initially, a computational approach is used to design a library of thousands of peptides that binds to the protein target. The peptides are sorted based on their predicted binding affinity, and hundreds of top-ranked potential candidates are used as probes for microarray screening and selection. The proposed combined approach was applied to identify peptides that bind to orexin A neuropeptide.
