Three-dimensional structures made of DNA hold the potential to encapsulate and release drugs, regulate the folding and activity of encapsulated proteins, selectively encage nanomaterials, and assemble 3D-networks for catalysis and biomolecule crystallization.  We will describe the synthesis of 3D-DNA structures, such as DNA cages and nanotubes, with deliberate variation of geometry, size, single- and double-stranded forms, permeability and length.  These can be readily switched to different sizes, and can be 'opened' or closed with added DNA strands.  The size selective encapsulation of nanomaterials within these nanostructures, and the release of this cargo when specific DNA strands are added will be described.  The incorporation of synthetic polymers, to create a new class of core-shell biohybrid materials will be shown.  Finally, we will describe the delivery of these responsive DNA cages and nanotubes into mammalian cells.
