Bis-(3'-5')-cyclic dimeric guanosine monophosphate (c-di-GMP) acts as a bacterial second messenger triggering the sessile-motile transition in bacteria. There is evidence that increasing concentrations of c-di-GMP promote biofilm formation in certain microbes. In some bacterial organisms, the signaling molecule nitric oxide (NO) can regulate biofilm aggregation and dispersal. In Shewanella woodyi (S. woodyi), a Heme-Nitric Oxide/Oxygen Binding domain (H-NOX) is in the same operon as a bi-functional diguanylate cyclase (DGC), suggesting a link between c-di-GMP turnover and NO sensing. Comparing wildtype and an h-nox gene disrupted mutant, we provide evidence NO/H-NOX regulates both c-di-GMP and biofilm formation of S. woodyi.
