Silybin, an active constituent of silymarin has been reported to show anti-cancer and anti-oxidant activities. However, it has poor water solubility and bioavailability. In this report, we describe the effect of �-cyclodextrin on the aqueous solubility and rate of dissolution of sylibin. The results of phase solubility studies show that the solubility of silybin is enhanced by the presence of �-cyclodextrin. The stoichiometry of the complexation was to be a 1:1 ratio, and the inclusion complex was found to be very stable. The complexes were prepared by four different methods: physical mixture, kneading, precipitation, and co-evaporation methods. The inclusion complex was characterized by FTIR, NMR, UV, differential scanning calorimetry, and SEM. Finally, the in-vitro studies indicate that the water solubility of silybin was greatly improved by the presence of �-cyclodextrin.
