Oxyhemoglobin I (Hb I) of the clam Lucina Pectinata is a monomeric protein that binds and transports hydrogen sulfide to the clam endosymbiotic sulfide-oxygen bacteria. HbI mutant was prepared using site directed mutagenesis.  Based on the results of previous observations, only 4 conditions of crystallization out of 50 formed crystals. Condition containing 1.5M Lithium Sulfate, 0.1M Na Hepes pH 7.5 managed to form single crystals after 24 hours.  A second and condition consisting of 28% PEG 400, 0.1Na Hepes pH 7.5, 0.2M Calcium Chloride and 20% iso-Propanol, 0.1M Na Acetate pH 4.6, 0.2M Calcium Chloride formed crystal plaques. The fourth condition with 30% MPD, 0.1M Na Acetate pH 4.6, 0.2M Calcium Chloride formed crystalline needle clusters. Further optimization HbI GluE7His mutant experiments are recommended to obtain crystals of better quality. Also, lowering some precipitant agent concentration may result in single crystals.
