The stringent sequence specificity of TEV protease makes it a very useful enzyme for cleaving fusion proteins.  This prompted us to alter the enzyme specificity parameters slightly to alter the active site structure in an attempt to create a novel specificity for these modified enzymes.  These experiments utilized the Green-Orange(G-O) fusion protein substrates developed in our lab to investigate and characterize the specificity of these TEV Protease mutants.  Additionally, we optimized the parameters required for storage and purification of G-O substrates and compared their fluorescent and kinetic parameters.
