Amphiphilic block copolymers can self-assemble into core-shell structured nanoparticles, which have been vastly investigated for in vivo drug delivery. Physical and chemical properties of nanoparticles greatly influence their pharmacokinetic behaviors which ultimately determine their capability of tumor accumulation and therapeutic efficacy.Herein, we report the synthesis of a series of amphiphilic block polymers PEG-polymethacrylates of SN38 (PEG-PolySN38) with varied PEG and polySN38 chain lengthes. A new process using one block copolymer to fabricate nanoparticles of different sizes was developed. The blood clearance and biodistribution were subsequently studies. The relationships of nanoparticles' pharmacokinetics with the overall nanoparticle size, the PEG and PolySN38 chain lengthes were then correlated.
