Nicotine, cocaine, methamphetamine, opiates, and alcohol abuse have been and still remain major public health problems. This address will highlight our studies directed toward the design and development of potential pharmacotherapies for treating drug abuse and addiction-related behaviors. The main focus will be on the development of the selective dopamine uptake inhibitor RTI-336, the selective kappa opioid receptor antagonist JDTic, and nicotine acetylcholine receptor (nAChR) ligands as well as monoclonal antibodies and vaccines. This address will also provide an overview of the design and development of [123I]RTI-55 (Dopascan, Iometopane) as a single photon emission computed tomography (SPECT) imaging ligand for use as a diagnostic agent for Parkinson's disease
