Transition metal ions such as RuIII and PtIV are able to oxidize guanine (G) derivatives through similar mechanisms.  Both RuIII and PtIV-bound G have highly positively polarized C8-H that can be deprotonated by OH- and undergo two-electron redox reaction. In this study reactions of [RuIII(NH3)5(Ino)] (Ino = Inosine) at high pH with O2 and without O2 were studied. Under aerobic conditions ruthenium 8-oxo-Ino was observed by Liquid Chromatography/Mass Spectrometry (LC/MS) and High Pressure Liquid Chromatography (HPLC). Evidence for the glycolysis of [RuIII(NH3)5(Ino)] into [RuIII(NH3)5(Hypo)] (Hypo = hypoxanthine) and ribose under anaerobic conditions was also observed by LC/MS and HPLC. We propose that [RuIII(NH3)5(Ino)] disproportionates to [RuII(NH3)5(Ino)] and [RuIV(NH3)5(Ino)] followed by sugar glycolysis of the RuIV species.
