Despite its history as a potent broad spectrum antimicrobial, the use of silver(I) in modern medicine is largely limited to the topical treatment of burns and other wounds. Due to the increasing occurrence of multi-drug resistant bacteria and the threat of weaponizable bacteria there has been a surge in research related to expanding the use of silver(I) beyond its typical mode of use. To date, our understanding of the bactericidal mechanism of silver(I) still remains incomplete, and only a handful of silver protein complexes have been deposited in the PDB. Structural elucidation of specific silver protein interactions or complexes that are either directly involved with or mediate bacterial cell death can give vital information needed to design more effective silver based drugs. The presented work describes the combination of X-ray crystallography and NMR for the structural characterization of lysozyme, azurin and a bacterial glutaredoxin in complex with silver(I).
