Sensitive bioassays, such as the E-screen and A-screen, allow for evaluation of runoff from livestock operations for compounds that elicit steroid hormone receptor responses. This study evaluated select runoff extracts that exhibited endocrine disrupting activity. One liter runoff sample extracts were processed by a fraction collection HPLC with ultraviolet (UV) and fluorescence (FL) detection. The fractions collected were then reanalyzed by the E-screen and A-screen bioassays to determine the fraction(s) responsible for the endocrine disrupting response. The fraction collector line was then moved to a tandem mass spectrometer (MS/MS), and the same extracts were analyzed by HPLC-UV-FL-MS/MS to identify peaks present in each fraction. Results from this analysis confirmed the main compounds, and presented possible unknown peaks, that are responsible for the bioassay activity. This technique may also be employed to identify polar contaminants as part of a toxicity indicator evaluation (TIE) testing scheme.
