Quantum dots (QD) are capable of reporting the local temperature based on the temperature-dependent changes in the excitonic emission characteristics, even at the single quantum dot level. Utilizing the QD nano thermometer, we recently demonstrated for the first time that the temperature responses inside living cells are heterogeneous. To improve the uncertainty of temperature measurements and to enable their wider-spread applications, we develop a glovebox-free method for preparing water-soluble and ratiometric temperature-sensitive Mn2+-doped QDs with a previously unreported base structure. We discuss how the temperature-dependent optical properties of the nanocrystals can be tuned by changing their structure and composition and potential applications.
