Athletes involved in highly competitive endurance sports have been known to cheat in order to enhance their performance. In response to this, various techniques for detecting the doping methods have been developed and implemented. However, autologous blood transfusions remain undetectable with the current analysis methods.  We are developing a capillary electrophoretic method through which autologous blood transfusions may be detected. To this end, the sample preparation plays as crucial as the separation method in obtaining results that can distinguish transfused red blood cells from native ones with fidelity and reproducibility. Suspension of the red blood cells, injection methods, separation buffer, and capillary diameter all play  significant roles in the outcome and reproducibility of the separation. We will present our development of the most effective method of preparation, suspension and separation of red blood cells for the identification of transfused red blood cells in a microliter volume sample.
