Microfluidic chip-based devices are expected to facilitate chemical analyses in terms of higher throughput and reduced analysis time. Porous monoliths have been used in applications including chromatographic separations and solid-phase extraction. This work focuses on the preparation of a methacrylate monolith inside of the cross section of the microchannel of the SU-8 CZE-ESI/MS microchip for the preconcentration and detection of test mixtures.We report the use of monolith prepared in situ by UV-initiated polymerization process that enables the formation of monoliths only within a specified space. The chromatographic behavior of this monolith for preconcentration or cleanup procedures was evaluated by both FL detection of rodamine 123 dye and CZE-ESI/MS analysis of drug mixture of nicotine and verapamil.In conclusion, this technique is a simple approach for the extraction, elution and detection of hydrophobic analytes without interruption or need for sample handling. Usage is not limited to the applications demonstrated here.
