Cell types ranging from bacterial pathogens to cancer cells display diverse carbohydrate overcoats that either aid or subvert human and animal immune reponses to these potentially lethal cells. Unfortunately, dissecting the roles of these sugars is complicated by the microheterogeneous mixtures and low relative abundances of particular structures found in the natural sources. In tribute to the excellent work carried out by the late Prof. Gin in dissecting the role of carbohydrates in the development of potent vaccine adjuvants, this talk will focus on the design and use of synthetic tools, including automated oligosaccharide synthesis and multivalent displays, to probe macrophage and other responses to pathogen-associated carbohydrates and will discuss the implications in designing better vaccine adjuvants.
