A data acquisition and processing methods that allows digitization, real-time analysis and calibration (DRAC) was developed and applied to beam-scanning second harmonic generation microscopy. In this method, digitization of the signal from a photomultiplier tube (PMT) occurs synchronously with the laser, capturing the peak voltage generated by each pulse. The laser repetition rate was doubled to 160 MHz a polarization rotating delay line. With four channels of 16-bit detection and a ~50% duty cycle, the data acquisition rate was ~610MBps. These data were processed during image acquisition on an graphics processing unit (GPU). Parallel processing on a GPU is ideal for real-time image analysis and allows flexibility in processing methods. Simple methods for counting and signal averaging were both performed to extend the linear dynamic range in these images. Because this method individually records each signal voltage, it retains all information necessary for self-consistent calibration of each image. Calibration algorithms can be rapidly executed on the GPU during image acquisition as often as required. The high data throughput and flexible processing afforded by DRAC will enable further development for rapid polarization modulation imaging.
