Self-assembled lipid bi-layers such as liposomes have been widely used as biomimetic models of cell membranes and as drug/gene delivery vehicles. Liposome encapsulation technique has been demonstrated to enhance their compatibility with the biological milieu in vitro and in vivo. Cell surface carbohydrates have specific interactions with proteins, which play an important role in various biological recognition processes such as fertilization, metastasis, inflammations and host�pathogen adhesion. Therefore, carbohydrate molecules serve as attractive molecules for surface modification of liposomes with purpose for tissues specific and biocompatibility. In this presentation, glyco-functionalized liposome systems were prepared by thin-film hydration and extrusion process followed by chemically selective glyco-functionalization of liposomes via Staudinger ligation. The size and stability of the liposomes were confirmed by dynamic light scattering (DLS) and the leakage of entrapped 5,6-carboxyfluorescein, respectively. Particularly, anchor lipid effects on the stability of glyco-functionalized liposomes were investigated by using two different anchor lipids namely (Cholesterol-PEG2000-TP) and (DSPE-PEG2000-TP) monitored via the fluorescence leakage assay. Furthermore, the density and accessibility of grafted carbohydrate residues on the liposome surface were evaluated.
