We have investigated organocatalytic strategies for the synthesis of Fmoc-protected �2- amino acids, which are useful building blocks of peptidomimetics. Our goal is to develop asymmetric methods that are compatible with the base-labile Fmoc-protecting group and result in the construction unnatural �-amino acids. Using enamine and SOMO catalysis, we have explored additions to Fmoc-protected 3-aminoproionaldehyde. The resulting enantioenriched products can be directly oxidized to the corresponding �2-amino acids.
