We report herein our recent findings on block co-polymer macromolecular amphiphiles synthesized using RAFT controlled radical polymerization.  Specific polymer-polymer interactions have been designed to increase the stability of polymer micelle assemblies, which we find yield high drug loading capacity for compounds that are known to be recalcitrant to packaging, such as camptothecin and 5-fluorouracil. Further, we have designed and synthetically installed molecular recognition sites to further maximize controlled drug loading and release.  These new polymer systems form drug-loaded nanoparticles in the sub 100 nm size regime and have been evaluated in biophysical context, cell culture as well as preliminary animal studies.
