The worldwide popular beverage beer is known to stimulate gastric acid secretion. However, the effects of different beer types or beer constituents have not been investigated. In the present study, the effects of different beers on mechanisms of gastric acid secretion were compared. To investigate compound specific effects, beer bitter acids and organic acids were quantified by UPLC-MS/MS and HPLC-DAD, respectively. The intracellular pH, as an indicator of proton secretion, was determined in HGT-1 cells. Furthermore, the expression of relevant genes, coding the H+/K+-ATPase, the receptors for histamine, acetylcholine (CHRM3) and somatostatin, was analyzed by qPCR. Besides succinic, malic and citric acid, also ethanol contributes to some extent to the effect of beer. We identified the bitter acids, classified as a-, �- and iso-a-acids, as potential key components promoting gastric acid secretion and up-regulating CHRM3 gene expression by a maximum factor of 2.01 with a correlation to their respective bitterness.
