DNA functionalized gold nanoparticles are being explored for the development of label free biosensors for detection of oligonucleotides. This detection is based on a colorimetric assay that utilizes complementary interactions of DNA and distance dependent optical properties of gold nanoparticles.  In this research we report the synthesis of a tripodal surface anchor, 2,4,9-Trithiaadamantane, modified with phosphoramidite end group from which specific oligonucleotide sequences were attached.  Due to the amphiphilic nature of DNA-capped tripod anchor, the stability of the GNPs is enhanced presenting a metal and organic hydrophobic core stabilized by hydrophilic DNA shell upon self-assembly of the ligand across the gold surface. Less susceptible to oxidation, heat and temperature fluctuations, this rigid, highly symmetric surface anchor has a large foot print providing 3:1 dilution per chain of DNA attached. This allows enough space for the colorimetric detection of association and dissociation of complementary sequences measured via temperature controlled UV-Vis spectroscopy.
