A new gemcitabine-poly(squalene methacrylate) (Gem-PSqMA) bioconjugate with tunable molar mass was synthesized by the RAFT technique. For the first time, stable PEGylated Sq-based nanoparticles were obtained by co-nanoprecipitation of squalene-poly(ethylene glycol) (Sq-PEG) and Gem-PSqMA. In vitro tests with nanoparticles of Gem-PSqMA on various cancer cell lines showed significant anticancer activity, sometimes even higher than that of the originally developed Gem-Sq nanoparticles. In addition, it appeared that the anticancer activity was governed by the polymer chain length.
