N-Modified phenylalanine (Phe) derivatives undergo self-assembly and hydrogelation in aqueous media. Side chain and C-terminal modifying groups exert a significant influence on the self-assembly and hydrogelation of Phe derivatives. Herein, we report the effect of various N-terminal modifying groups, including aromatic and nonaromatic functionality, on the self-assembly/hydrogelation of Phe-derived amino acids. It was found that aromatic functionality at the N-terminus was more effective at promoting hydrogelation than nonaromatic functionality and that aromatic surface area affects hydrogel formation/mechanical rigidity. These empirical studies illuminate the influence of N-terminal hydrophobicity and structural architecture on efficient self-assembly and hydrogelation.
