The biological application of nanoparticles has generated increased need for robust, accurate, and precise methods for characterizing their molar mass, size, shape, and compactness. We showcase the power of hydrodynamic chromatography (HDC) coupled on-line to multi-angle static and quasi-elastic light scattering, differential viscometry, and differential refractometry to characterize a nanosized, cage-like ('nanocage') drug delivery vesicle. Using the quadruple-detector HDC method, the absolute molar mass, various sizing parameters, and the shape and structure of the nanocage were all determined continuously and as a function of HDC elution volume. This was achieved in less than twenty minutes, a fraction of the time needed for most other types of analysis, and provided a more complete physical picture of the analyte than that obtained using traditional techniques.
