Pyocyanin is a redox active secretory product of Pseudomonas aeruginosa which is a bacterium that frequently causes acute pneumonia. The activation of pyocyanin is a potential source of the oxidative damage caused by P. aeruginosa during infection in mammalian cells. In this paper, a commercial electrochemical platform was coupled with LC-MS to evaluate the redox activity of pyocyanin in the scanning mode and facilitate detection in the direct current mode. Using both glassy carbon and Magic diamond working electrodes, redox profiles were collected during pyocyanin infusion in tandem with the mass spectrum. Oxidative products were confirmed using exact mass on a time of flight MS system. The system was switched into DC mode and spiked human serum samples were tested after C18 SPE processing. We observed the major oxidative product on the LC-EC-MS system down to spiked levels of 50 ng/mL testing only 200 �L of seurm.
