We confirmed that linear calibration curves for quantifying unconjugated estriol (uE3), alpha-fetoprotein (AFP), and human chorionic gonadotropin (hCG) using competitive and sandwich 1,1'-oxalyldiimidazole chemiluminescent enzyme immunoassays (ODI CLEIAs) in the presence of Triton X-100 (0.1 wt % ) are multiple times wider than those in the absence of Triton X-100. The wider linear dynamic range results from Triton X-100 acting as an interference in the interaction between the hydrophobic substrate and the hydrophilic enzyme (e.g., horseradish peroxidase (HRP), alkaline phosphatase (ALP)) to produce a fluorescent dye. These results indicate that rapid quantification of AFP, uE3, and hCG using cost effective and highly sensitive ODI CLEIAs in the presence of Triton X-100 can be applied as an accurate, precise, and reproducible method to diagnose genetic disorders (e.g., trisomy 18 and trisomy 21) in fetuses.
