Cell penetrating peptides, or protein transduction domains, are promising agents for the intracellular delivery of large, hydrophilic molecules which are unable to pass the plasma membrane by themselves. Among the protein transduction domains, arginine-rich peptides are the most efficient subclass. In the light of this information, we designed and synthesized protein transduction domain mimics which contain guanidinium functionality along the polyoxanorbornene backbone. Structure activity relationship has been studied with designed molecules in order to be able to determine the requirements for efficient cellular internalization, as well as the exceptional activity in delivery of bioactive molecules such as siRNA and proteins has been demonstrated
